Description

The contact-mediated signaling is a general mechanism for morphological differentiation of many cells and is not unique to cells of neuronal origin only. In multicellular organisms, the signaling proteins (morphogens) move from signaling centers where they are produced to target cells whose growth and patterning they regulate. In fruit fly, one of the morphogen identified is transforming growth factors-beta family member decapentaplegic (Dpp) which is produced in wing imaginal disc and is involved in long-distance signaling. Cells that receive Dpp and activated Dpp signal transduction extend cytonemes that directly make contact with the Dpp-producing cells (1). The contact mediated signaling via cytonemes is also a hallmark for diaphanous, neuroglian, capricious and Shibire (1). Mutations in the fruit fly Shibire gene led to several temperature sensitive mutants, at non permissive temperature the flies are paralyzed and have defect in endocytotic transport and vesicle recycling (2). The shibire mutation also affects many tissues outside the nervous system. One of the product of Shibire gene is Dynamin which is expressed ubiquitously and associated with membrane fraction. The Drosophila melanogaster Dynamin has high homology to mammalian Dynamin proteins.

Dynamin is a GTP-driven mechanochemical enzyme homologues to yeast mx-protein. Dynamin family of proteins are member of GTPases. The members of the Dynamin family include Dynamin I, Dynamin II and Dynamin III, which are microtubule associated proteins which are involved in endocytosis, synaptic transmission and neurogenesis. The Dynamin I expression is localized in brain, Dynamin II is ubiquitous with highest expression in testis and Dynamin III transcripts are found in brain (2). Dynamin-1 is a microtubule-associated force-producing protein involved in producing microtubule bundles and able to bind and hydrolyze GTP.
